Confocal principle
Perform 3D imaging on the entire sample
The LSM800 laser confocal microscope captures the defined light sections in the sample using the laser in the confocal beam path and combines them in a three-dimensional image. Its aperture (usually referred to as a pinhole) is set in such a way that information outside the focal length is blocked, and only information within the focal length can be detected.
·Scan and generate images along the x and y axes. Information within the focal length is bright, while information outside the focal length is dark.
·Generate an image stack by changing the distance between the sample and the objective lens (the sample is an optically cut section).
·By analyzing the distribution intensity of individual pixels in the image stack, the corresponding height can be calculated. The height information in the entire field of view can be combined to form a height map.
